The most important biocontrol agents are considered entomopathogenic nematode. Due to growing application of this pathogen in recent decades, the identification of these factors is very important. The study in 2011 from three different habitats were sampled of South Island.
In continuing efforts to morphological characteristics, molecular and crossbreeding tests of native species collected are used for identification. With galleria trap method, of 60 soil samples collected, five samples of the two Steinernemaand Heterorhabditisgenera was isolated. Determining classic specification according to apparent characteristics and morphometricthe grouping so called isolates Boj1, Boj7, Boj8 and Boj9 group "feltiae" and HBoj isolates was in group "bacteriophora". The phylogenetic analysis on sequence data, gene regions ITS and 28S, showed that isolates attributed to Boj1, Boj7, Boj8 and Boj9 belonging to the "feltiae" group "Steinernema" genera and also the HBoj isolates was belonging to "bacteriophora" group of "Heterorhabditis" genera. ). Reconstruction of phylogenetic trees using both ITS and D2/D3 genes showed that four isolates, Boj1, Boj7, Boj8 and Boj9 were grouped in a clade with other species of "feltiae"group (of Steinernema)These results were also verified in reconstructed phylogenetic tree for "bacteriophora" group species and HBoj isolate based on ITS gene.
Cross breeding tests result between species and Boj1, Boj7, Boj8 and Boj9 isolates with S.feltiaereference isolates, it was confirmed that the first isolates (Boj1) belongs to the S.feltiaespecies.
The other three isolates (Boj7, Boj8 and Boj9), was awarded the "Steinernema" genus Online survey, the first study of entomopathogenic nematode native population in North Khorasan.
